If I am mentioning DmC: Devil May Cry, I would italicize the whole title, but if I am referring to Call of Duty as CoD or The Last of Us as TLoU, would those abbreviations be italicized?

Why would you italicize one title and not another? Your examples are inconsistent. What makes DMC (or, strangely, DmC in your example—but that's another issue) any different than COD (or COD)?

There's no general rule for italicising titles: it's simply a matter of style. If you're required to follow a particular style manual, check what it says. As Jason says, make sure you're consistent in your approach. NB if you pronounce each letter of a word (e.g. FBI or CIA) it's an initialism, vs an acronym where you pronounce the whole word, e.g. NATO, GIF, scuba. It's usual for all letters in an initialism to be all-caps. Wikipedia has a useful page on this. :-)

I decided to do a brief check for some four letter abbreviations at forums I visit just to verify that, and if you are wondering why I need four letter abbreviations, it is to bypass the minimal character limit search restriction.
Typically, I only see this sort of abbreviation on websites, and when do, I do not observe these being given special treatment. This is even true of The Battle of Wesnoth. People usually just write out "HttT" which is the popular abbreviated form for Heir to the Throne campaign. If anybody had a vested interest in presenting this type of abbreviation in good form, it would be them, because the community is actively developing the game on that website, and it is probably one of the most popular G.P.L. licensed games. Similarly doing a brief check on google to see how people treat "LttP", which is the popular way of abbreviating A Link to the Past it does not seem like people italicize that either. A brief check on the Caves of Narshe verifies that people do not bother to italicize "LttP" and its variations either. Immaterial and Missing Power is popularly abbreviated to "IaMP" in Touhou communities, and I do no not remember seeing that italicized either (example).
